اثر مصرف حاد محلول خوراکی عناب قبل از یک جلسه ورزش مقاومتی دایره‌ای بر آپوپتوز نوتروفیل انسانی

چکیده

سابقه و هدف. مطالعات اندکی در خصوص ورزش و مکمل‌گیری و آثر آن بر آپوپتوز نوتروفیل وجود دارد. اما درباره تمرین مقاومتی و مکمل‌گیری عناب به عنوان یک گیاه دارویی سنتی شواهدی یافت نشد. لذا، هدف از تحقیق حاضر اثر مصرف حاد محلول خوراکی عناب یک ساعت قبل از یک جلسه ورزش مقاومتی دایره‌ای بر آپوپتوز نوتروفیل انسانی بود.
مواد و روش‌ها. تعداد 14 دانشجوی داوطلب پسر جوان بطور تصادفی به دو گروه 7 نفره شامل گروه تمرین مقاومتی دایره‌ای با دارونما  و گروه تمرین مقاومتی دایره‌ای با محلول عناب  تقسیم شدند؛ و یک ورزش مقاومتی دایره‌ای را انجام دادند. آزمون شامل سه دایره بدون توقف با 3 دقیقه استراحت فعال بین دورها، و هر حرکت 30 ثانیه (حدود 14- 10 تکرار) با 75% یک تکرار بیشینه (1RM) بود. افراد یک ساعت قبل از ورزش دارونما و محلول عناب (0.5 گرم به ازای هر کیلوگرم وزن بدن در 2.5 سی‌سی آب مقطر) را به صورت دوسوکور دریافت نمودند. نمونه‌های خونی یک ساعت قبل از مصرف محلول عناب، بلافاصله و 2 ساعت پس از ورزش جمع‌آوری گردید. پس از جداسازی و شمارش نوتروفیل‌ها، آپوپتوز نوتروفیل توسط کیت AnexinV-FITC و روش فلوسایتومتری اندازه‌گیری شد.
یافته‌ها. در گروه محلول عناب کاهش معناداری در نوتروفیل‌های آپوپتوزی اولیه مشاهده شد ولیکن افزایش معناداری در گروه دارونما یافت شد (0.048 = p). از طرف دیگر نوتروفیل‌های آپوپتوزی تأخیری در گروه محلول عناب تغییر معناداری نکرد، اما در گروه دارونما کاهش معناداری مشاهده شد (0.003 = p).
نتیجه‌گیری. عناب حاوی کربوهیدارت‌ها و اسیدهای آمینه‌ای نظیر آلانین، اسید آسپارتیک، و مخصوصاً اسید گلوتامیک می‌باشد که می‌توانند در شکل‌گیری گلوتامین در بدن شرکت کنند. از این رو می‌توان نتیجه گیری نمود که مهار آپوپتوز در گروه عناب شاید به دلیل فراهمی گلوکز و گلوتامین بدن در نتیجه فراهمی آن‌ها یک ساعت قبل از آزمون توسط مصرف محلول عناب بوده باشد.

Background & Purpose: there are a few studies about effects of jujube extraction and exercise status on nutrophils. The aim of present research was the acute effect of jujube solution an hour before circuit resistance exercise on human neutrophil apoptosis.
Methodology: Fourteen young male volunteer students were divided into two groups of placebo and jujube solution randomly, performing one circuit resistance exercise (9 moves/stops, 30 seconds for each exercise, 3 nonstop sets with a 3-minute active recession between sets, 10-14 repetitions, and an intensity of 75% one repeat maximum). Subjects received placebo and jujube solutions (0.5 g/kg body weight in 2.5cc of distilled water) an hour before test. Blood samples were collected 60 minutes before feeding, immediately, and 2 hours after the exercise for separation and counting the number of neutrophils, and neutrophil apoptosis was determined through AnexinV-FITC kit and flow cytometric method.
Results: Early Apoptotic Cells decreased in response to circuit resistance exercise in group of Jujube Solution feeding, and increased in Placebo group. Delayed Apoptotic Cells unchanged in response to circuit resistance exercise in group of Jujube Solution feeding but decreased in Placebo group.
Conclusion: Therefore, it might be stated that loading with edible jujube solution can be effective in inhibiting apoptosis at least for one hour before exercise. The present findings indicate that perhaps useful effects of jujube can be explained by existing glucose compounds and amino acids which could provide neutrophils with proper nutrition source in comparison with placebo.
