اثر هشت هفته تمرین استقامتی و مصرف عصاره سندانی بر شاخص های عملکردی کبد و کلیه در موشهای تغذیه شده با رژیم غذایی پرکلسترول

چکیده

زمینه و هدف: چاقی و عوارض ناشی از آن نظیر پرفشاری خون، کبد چرب، دیابت نوع 2 و نارسایی‌های کلیوی از موارد اصلی تهدید‌کننده حیات تلقی می‌گردند. از اینرو هدف از این مطالعه اثر هشت هفته تمرین استقامتی و مصرف عصاره سندانی بر شاخص­های عملکردی کبد و کلیه در موشهای صحرایی تغذیه شده با غذای پرکلسترول بود.
مواد و روش‌ها: 35 سر موش صحرایی نر نژاد ویستار به صورت تصادفی به پنج گروه هفت تایی؛ کنترل نرمال (CON)، کنترل هیپرکلسترولمی (HC)، عصاره الکلی گیاه سندانی (Ext)، تمرین (E) و تمرین همراه با عصاره سندانی  (Ext+E)، تقسیم شدند. عصاره گیاه سندانی به مقدار 100 میلی گرم به ازای هر کیلوگرم وزن موش ترکیب و سه روز در هفته، بمدت 8 هفته بصورت گاواژ قبل از تمرین به موش ها داده شد. گروه­های تمرین،به مدت هشت هفته هر هفته 5 روز و هر روز برای 30 دقیقه دویدن روی نوار گردان را با شدت 70-65 درصد VO2max انجام دادند. متغیرهای بیوشیمیایی آلانین آمینو ترانسفراز(ALT)،آسپارتات آمینو ترانسفراز (AST)، آلکالن فسفاتاز (ALP)، کراتین، آلبومین، بیلی روبین و پروتئین تام سرم خون، 24 ساعت پس از آخرین جلسه تمرینی مورد سنجش قرار گرفت.
نتایج: نتایج آزمون تحلیل واریانس یک طرفه نشان داد بین گروهها اختلاف معنی‌داری بین میزان کلسترول، AST، ALT ، BUN، کراتین ، آلبومین،بیلی روبین و پروتئین تام وجود دارد (05/0P≤). نتایج آزمون تعقیبی توکی نشان داد در مقادیر AST بین گروه کنترل هایپرکلسترومی با گروه تمرین (026/0=p) تفاوت معناداری وجود دارد. همچنین بین گروه تمرین با تمرین+عصاره نیز تفاوت معناداری وجود داشت (027/0=p). میزان ALT در گروه تمرین+عصاره نسبت به گروه تمرین (006/0=p) و نسبت به کنترل هایپرکلسترومی (0001/0=p) بطور معناداری افزایش یافته بود. مقادیر پروتئین تام در گروه تمرین نسبت به گروه کنترل هایپرکلسترومی(011/0=p) و تمرین+عصاره نسبت به گروه کنترل هایپرکلسترومی (0001/0=p) بطور معناداری افزایش یافته بود. میزان آلبومین و بیلی روبین و کراتین در گروه عصاره+تمرین نسبت به کنترل هیپرکلسترولمی بطور معناداری بالاتر بود (05/0p≤).
نتیجه‌گیری: : یافته های تحقیق حاضر نشان داد شیوه تمرینی و مقدار عصاره سندانی به کار رفته در این تحقیق میزان عملکرد کلیوی و کبدی  را تحت تأثیر قرار نمی دهد. پیشنهاد می شود در مطالعات آینده دوزهای متفاوت این گیاه مورد بررسی قرار گیرد.

The effect of endurance training and Anvillea garcinii extract consumption on liver and kidney functional indices in rats fed with high cholesterol diet

Background & Purpose: Obesity and its complications such as high blood pressure, fatty liver, type 2 diabetes and kidney failure are considered to be the main life-threatening cases. Therefore, the aim of this study was to investigate of eight weeks of endurance training and consumption of Anvillea garcinii extract on liver and kidney functional indices in rats fed with high cholesterol diet.
Methodology: 35 male Wistar rats randomly divided into five groups of seven; Normal control (CON), hypercholesterolemic control (HC), alcoholic extract of anvil plant (Ext), exercise training (E) and exercise training combined with anvil extract (Ext+E). The extract of anvil plant in the amount of 100 mg per kilogram of the weight of the rat was given by gavage three days a week for 8 weeks. The training groups ran for 30 minutes on the treadmill for 5 days a week for eight weeks with an intensity of 65-70% of VO2max. The biochemical variables of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), and creatinine, albumin, and bilirubin and total protein of blood serum were measured 24 hours after the last training session.
Results: The results of the one-way analysis of variance test showed that there is a significant difference between the groups between the levels of cholesterol, AST, ALT, bilirubin, creatinine, albumin, bilirubin and total protein (P≤0.05). The results of Tukey's post hoc test showed that there is a significant difference in AST values ​​between the (HC) group and the (E) group (p=0.026). Also, there was a significant difference between the (E) group and (Ext+E) (p=0.027). The amount of ALT in (Ext+E) group was significantly increased compared to (E) group (p=0.006) and compared to (HC) group (p=0.0001). The amount of total protein in the (E) and (Ext+E) groups compared to the (HC) group were significantly increased (p≤0.05). The amount of albumin, bilirubin, and creatinine in the (Ext+E) group was significantly higher than the (HC) (p≤0.05).
Conclusion: The findings of this research showed that the training method and the amount of anvil extract used in this research does not affect the level of kidney and liver function. It is suggested to investigate different doses of this plant in future studies.
