تأثیر تمرین هوازی بر بیان ژنIL-10 و TNF-α در بافت کبد موش‌های صحرایی فاقد تخمدان و تغذیه شده با غذای پرچرب

چکیده

اهداف: هدف اصلی این مطالعه تأثیر تمرین هوازی بر بیان ژن اینترلوکین-10 (IL-10) و  فاکتور نکروز دهنده توموری آلفا (TNFα) در بافت کبد موش­های صحرایی فاقد تخمدان و تغذیه شده با غذای پرچرب بود.
روش مطالعه: سی و دو سر موش صحرایی ماده به طور تصادفی به چهار گروه 1)کنترل نرمال، 2) اوارکتومی+ مصرف غذای پرچرب، 3) اوارکتومی+ مصرف غذای پرچرب+ تمرین با شدت متوسط، 4) اوارکتومی+مصرف غذای پرچرب+ تمرین شدید، تقسیم شدند. موش­­های گروه تمرین به مدت 8 هفته و هر هفته 5 جلسه تمرین هوازی شدید  و یا با شدت متوسط را روی نوارگردان انجام دادند. 48 ساعت پس از آخرین جلسه تمرین، بافت کبد به منظور ارزیابی بیان ژن  IL-10  و TNFα به روش Real Time PCR برداشت شد. همچنین سطوح سرمی آنزیم­های آلانین ترانس آمیناز (ALT) و آسپارتات آمینوترانسفراز (AST) اندازه­گیری شد.
یافته­ ها: افزایش معنادار بیان ژن TNFα (003/0= p) و کاهش بیان ژن IL-10 کبد در گروه اوارکتومی+ مصرف غذای پرچرب نسبت به گروه کنترل نرمال مشاهده گردید (01/0=p). در هر دو گروه تمرین با شدت متوسط و بالا بیان ژن TNFα نسبت به گروه اوارکتومی+ مصرف غذای پرچرب بطور معناداری پایین­تر بود (001/0= p) اما مقادیر بیان ژن IL-10 در دو گروه تمرین با شدت متوسط و بالا نسبت به گروه اوارکتومی+ مصرف غذای پرچرب تفاوت معناداری نداشت. همچنین تفاوت معناداری در سطوح سرمی دو آنزیم AST و ALT بین گروه­های پژوهش یافت نشد.
نتیجه گیری: یافته­های مطالعه حاضر نشان می­دهد تخمدان برداری و مصرف غذای پرچرب موجب ایجاد وضعیت التهابی در کبد می­شود. تمرینات هوازی با شدت متوسط و بالا در بهبود این وضعیت اثرگذار است.

Objectives: The main objective of this study was to investigate the effect of aerobic training on the expression of interlukin-10 (IL-10) and tumor necrosis factor-alpha (TNFα) genes in the liver tissue of ovariectomized and high-fat diet fed rats.
Methods: Thirty-two female rats were randomly divided into four groups: 1) Normal control, 2) Ovariectomy + high-fat diet, 3) Ovariectomy + high-fat diet + moderate-intensity training, 4) Ovariectomy + high-fat diet + high-intensity training. The training groups underwent aerobic training sessions for 8 weeks, five sessions per week, either at a moderate intensity or at a high intensity on a treadmill. Liver tissue samples were collected 48 hours after the last exercise session to evaluate the expression of IL-10 and TNFα genes using the Real-Time PCR method. Serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) enzymes were also measured.
 Results: Significant increase in TNFα gene expression (p=0.003) and a decrease in IL-10 gene expression (p=0.01) in the liver were observed in the ovariectomy + high-fat diet group compared to the normal control group. In both moderate and high-intensity training groups, TNFα gene expression was significantly lower than in the ovariectomy + high-fat diet group (p=0.001), but there was no significant difference in IL-10 gene expression values ​​in the moderate and high-intensity training groups compared to the ovariectomy + high-fat diet group. Also, no significant difference was found in the serum levels of the two enzymes AST and ALT between the study groups.
Conclusion: The findings of the present study indicate that ovariectomy and high-fat diet consumption cause an inflammatory state in the liver. Moderate and high intensity aerobic training is effective in improving this condition.
