بررسی اثر تمرین هوازی بر بیان ژن شاخص رشد اندوتلیال عروق- (VEGF-B) Bو وضعیت آنتی اکسیدانی تام (TAS) بافت قلب در موش‌های صحرایی دیابتی

چکیده

سابقه و هدف: دیابت با هایپرگلایسمی و افزایش استرس اکسیداتیو باعث اختلالات قلبی و عروقی شده، بدین ترتیب سبب بروز بیماری­های شریان کرونری یا پرفشارخونی سیستمیک می­شود. از این رو هدف از مطالعه اخیر بررسی اثر تمرین هوازی بر بیان ژن شاخص رشد اندوتلیال عروق-B (VEGF-B) و وضعیت آنتی اکسیدانی تام (TAS) بافت قلب در موش­های صحرایی نر دیابتی بود.
مواد و روشها: پژوهش حاضر از نوع تجربی است و در آن 30 سر موش صحرایی نر بالغ به 3 گروه 10 تایی شامل گروه دیابت تمرین، دیابت کنترل و سالم کنترل تقسیم شدند. موش­های گروه­های دیابتی از طریق تزریق درون صفاقی استرپتوزوتوسینmg/kg)  45 (دیابتی شدند. برنامه تمرینی ورزشی شامل 6 هفته تمرین روی نوارگردان بود. 24 پس از ساعت از آخرین جلسه تمرین برای بررسی اثر تمرین، نمونه­های بافت قلب به منظور اندازه گیری بیان ژن شاخص رشد اندوتلیال عروق-B و TAS استخراج گردید. برای مقایسه بین گروهی و اختلاف معنادار بین گروه­ها از تحلیل واریانس یک طرفه و آزمون تعقیبی LSD با سطح آماری (05/0˂P) استفاده گردید.
یافته‌ها: یافته­ ها نشان داد که پس از یک دوره تمرین استقامتی میزان بیان ژن VEGF-B و TAS در گروه دیابت تمرین نسبت به دیابت کنترل افزایش معنادار داشته است (001/0P=)، اما میزان بیان ژن VEGF-B و TAS بین دو گروه دیابت تمرین و سالم کنترل به ترتیب عدم تفاوت معنادار (364/0P=) و تفاوت معناداری (033/0P=) مشاهده شد.
نتیجه گیری: به نظر می­رسد تمرین استقامتی می­تواند اثر مثبت بر میزان بیان ژن VEGF-B و سطح TAS بافت قلب موش­های دیابتی داشته باشد و می­تواند باعث بهبود قلب دیابتی شود.

چکیده [English]

Background and Purpose: Diabetes causes cardiovascular disorders and coronary artery disease or systemic hypertension. Therefore, the aim of the present study was to evaluate the effect of aerobic exercise on vascular endothelial growth factor-B (VEGF-B) gene expression and the amount of total antioxidant status (TAS) of cardiac tissue in diabetic male rats.
Methodology: This experimental study was performed on 30 adult male Wistar rats and divided into 3 groups of Diabetic training, Diabetic control and healthy control (n = 10). Diabetes was induced by a single dose of streptozotocin (45 mg/kg). The exercise program consisted of 6 weeks of treadmill training. 24 hours after of the last training session, cardiac tissue samples were extracted to measure VEGF-B gene expression and TAS. One-way ANOVA and LSD test with statistical level of (P˂0.05) were used to compare between groups differences.
Results: Results showed that after one period of endurance training there was a significant increase in VEGF-B gene expression and TAS in the DT group compared to CD group (P = 0.001). There was no significant difference (P = 0.364) in VEGF-B gene expression, also a significant difference (P = 0.033) was in antioxidant status in DT and HC groups.
Conclusion: It seems that endurance training has a positive effect on VEGF-B and TAS of in the heart of diabetic rats.
