Abstract
Objectives: The main objective of this study was to investigate the effect of aerobic training on the expression of interlukin-10 (IL-10) and tumor necrosis factor-alpha (TNFα) genes in the liver tissue of ovariectomized and high-fat diet fed rats. Methods: Thirty-two female rats were randomly divided into four groups: 1) Normal control, 2) Ovariectomy + high-fat diet, 3) Ovariectomy + high-fat diet + moderate-intensity training, 4) Ovariectomy + high-fat diet + high-intensity training. The training groups underwent aerobic training sessions for 8 weeks, five sessions per week, either at a moderate intensity or at a high intensity on a treadmill. Liver tissue samples were collected 48 hours after the last exercise session to evaluate the expression of IL-10 and TNFα genes using the Real-Time PCR method. Serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) enzymes were also measured. Results: Significant increase in TNFα gene expression (p=0.003) and a decrease in IL-10 gene expression (p=0.01) in the liver were observed in the ovariectomy + high-fat diet group compared to the normal control group. In both moderate and high-intensity training groups, TNFα gene expression was significantly lower than in the ovariectomy + high-fat diet group (p=0.001), but there was no significant difference in IL-10 gene expression values in the moderate and high-intensity training groups compared to the ovariectomy + high-fat diet group. Also, no significant difference was found in the serum levels of the two enzymes AST and ALT between the study groups. Conclusion: The findings of the present study indicate that ovariectomy and high-fat diet consumption cause an inflammatory state in the liver. Moderate and high intensity aerobic training is effective in improving this condition.
